Principle of the Assay::The Human TSLP ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human TSLP in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human TSLP coated on a 96-well plate. Standards and samples are pipetted into the wells and TSLP present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human TSLP antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TSLP bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Thymic stromal lymphopoietin, also called TSLP is a protein belonging to the cytokine family. This gene is mapped to 5q22.1. It encodes a hemopoietic cytokine proposed to signal through a heterodimeric receptor complex composed of the thymic stromal lymphopoietin receptor and the IL-7R alpha chain. It mainly impacts myeloid cells and induces the release of T cell-attracting chemokines from monocytes and enhances the maturation of CD11c (+) dendritic cells. The protein promotes T helper type 2 (TH2) cell responses that are associated with immunity in various inflammatory diseases, including asthma, allergic inflammation and chronic obstructive pulmonary disease. The protein is therefore considered a potential therapeutic target for the treatment of such diseases
